Engine numbers typically 0 to 30 out from the frame. Engine always seemed to be the higher number (if not a match).

Location on the engine?? On top of the crankcase RHS behind the cylinders. Underneath the RH carb. There is a big cast flat spot and this has the Number stamped into it.
For changes what do you mean- typically they would increase by one for every engine?

Generally... 2000020 ~ 2020270 is the CB400N and CB250N 2100005 ~ 2124015 is the CB400NA and CB250NA 2200010 ~ 2210382 is the CB400NB and CB250NB 2300001 ~ 2310205 is the CB400NC and CB250N-DC ND MODELS 240****** 230 on, probably has the external oil feeds to the camshaft Earlier models have captive cam-chain blades later models don't. One of the front mounting bolts on the earlier models is an M10, and becomes M8, on later models. Etc Etc......but most of engines are very similar. Depends what your trying to find???
