i am not sure if i have asked this question before but is it possible to use one caliper from a 400 s/d on a 250s/d? or are they different sizes?

Remember - the single disc on the 250 was a bigger affair.

To use on the CB250N with it's 276mm Disc...
The CB400N single pot caliper would have to be fitted with a CB750 /CB900F Caliper hanger bracket.
The CB400NC twin pot would have to be fitted with an XBR 500 caliper bracket.

The CX500 also has 240mm discs I think, so would need the CB750F/CB900F hanger . To put a single 240mm disc and Nissan Caliper on the CB250N would only give the braking capacity of the CB125. As the CB250N has only adequate braking with the 276mm disc !
